      目的:观察姜黄素对肥胖SD大鼠体质量、血糖、血脂、葡萄糖耐量和脂肪组织炎症因子巨噬细胞趋化蛋白-1(monocyte chemoattractant protein-1,MCP-1)及肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)表达和分泌的影响。方法:将雄性SD大鼠分别用对照饲料(control diet,CD)和高脂饲料(high fat diet,HFD)喂养9周,HFD组每天给予低、中和高剂量姜黄素(10、30、100 mg/kg),共计3周。观察姜黄素对葡萄糖耐量、体质量和血清生化参数的影响。采用实时荧光定量PCR检测脂肪组织MCP-1及TNF-α mRNA的表达。结果:高剂量的姜黄素能明显改善HFD诱发的糖耐量受损(GTT实验15 min和30 min血糖,HFD对照组分别为:(19.4±1.7) mmol/L和(16.2±1.3) mmol/L;HFD高剂量姜黄素组分别为:(13.8±1.4) mmol/L和(12.1±0.9) mmol/L;P均小于0.05。中和高剂量姜黄素处理能降低HFD动物的体质量(F=13.219,P<0.001),3个剂量组姜黄素均能降低脂肪组织MCP-1(F=7.024,P=0.001)和TNF-α mRNA(F=5.537,P=0.004)表达,血脂成分和血糖有不同程度的改善,改善程度与使用的剂量有关。结论:姜黄素能降低肥胖SD大鼠脂肪组织MCP-1和TNF-α的基因表达,从而降低脂肪组织炎症因子的产生,可能是姜黄素改善高脂饮食诱发的代谢紊乱的作用机理之一,且高和中剂量组的改善作用优于低剂量组。

      Objectives:To examine the effects of curcumin on body weight,blood glucose,blood lipid,glucose tolerance and monocyte chemoattractant protein 1(MCP-1) and tumor necrosis factor-α(TNF-α) expression and secretion. Methods:Male SD rats were fed with either high fat diet(HFD) or control diet(CD). HFD-induced obese rats were randomly divided into 4 groups:HFD with saline treatment,HFD plus 10 mg/kg,30 mg/kg,and 100mg/kg of curcumin,respectively,with 10 rats in each group. Curcumin was applied to obese rats for 3 weeks. Body weight in each group was monitored weekly,and glucose tolerance test was performed. Serum lipid and glucose levels were detected using an automated biochemistry analyzer. The transcriptional levels of MCP-1 and TNF-α in epidi-dymis white adipose tissues(WAT) were examined by real-time PCR. Results:Three weeks after the treatment with curcumin,body weight and the expression of MCP-1 and TNF-α mRNA of the obese rats were significantly decreased. The contents of serum lipids and glucose were also improved and high and middle dose groups were better than low dose group in improving HFD induced metabolic dysfunction. Conclusion:Curcumin treatment can improve metabolic dysfunction induced by HFD,possibly through an inhibitory effect on the expression of inflammatory cytokines in WAT in obese rats. In addition,high and middle doses of curcumin exhibit better effect than low dose of curcumin.
